The role of neuropilin 2 in physiological and pathological angiogenesis

چکیده
Neuropilin 2 (NRP2) is a transmembrane receptor protein that was first discovered on neurons and then endothelial cells. On endothelial cells, it serves as co-receptor with the vascular endothelial growth factor receptor (VEGFR) to bind VEGF and induce a pro-(lymph)angiogenic intracellular signal. In addition to VEGF, NRP2 is also a receptor for semaphorin 3F (SEMA3F), which upon binding to NRP2 and Plexin A1 induces a strong anti-angiogenic signal. It is our hypothesis that SEMA3F could be a potentially effective treatment for metastatic cancers. A greater understanding of the regulation and expression of its receptor, NRP2, is needed. While NRP2 has been most robustly studied for its role in the vasculature, recent studies have shown that it is expressed on other cell types as well such as dendritic cells, T-cells, and visceral smooth muscle cells. In this study, we used western blot and immunohistochemistry to explore various different organs and cell types in an attempt to locate other novel locations of NRP2 expression. In particular, we found several new tissues that express NRP2

BOSTON UNIVERSITY SCHOOL OF MEDICINE Thesis THE ROLE OF NEUROPILIN 2 IN PHYSIOLOGICAL AND PATHOLOGICAL ANGIOGENESIS AND LYMPHANGIOGENESIS
The generation of new lymphatic vessels through lymphangiogenesis has been implicated in many disease states. This process has some overlap with the better studied angiogenesis pathway, but is under distinct molecular control. Specifically, it has been shown that VEGFR-3 and neuropilin-2 are important mediators of lymphangiogenesis. A greater understanding of this process could lead to new ther...
متن کاملTargeting Neuropilin-1 to Inhibit VEGF Signaling in Cancer: Comparison of Therapeutic Approaches
Angiogenesis (neovascularization) plays a crucial role in a variety of physiological and pathological conditions including cancer, cardiovascular disease, and wound healing. Vascular endothelial growth factor (VEGF) is a critical regulator of angiogenesis. Multiple VEGF receptors are expressed on endothelial cells, including signaling receptor tyrosine kinases (VEGFR1 and VEGFR2) and the nonsig...

متن کاملIn vitro combination therapy of pathologic angiogenesis using anti-vascular endothelial growth factor and anti-neuropilin-1 nanobodies
Objective(s): Emergence of resistant tumor cells to the current therapeutics is the main hindrance in cancer treatment. Combination therapy, which mixes two or more drugs, is a way to overcome resistant problems of cancer cells to current treatments. Nanobodies are promising tools in cancer therapy due to their high affinity as well as high penetration to tumor sites....
متن کاملGlycosaminoglycan modification of neuropilin-1 modulates VEGFR2 signaling.
Neuropilin-1 (NRP1) is a co-receptor for vascular endothelial growth factor (VEGF) that enhances the angiogenic signals cooperatively with VEGFR2. VEGF signaling is essential for physiological and pathological angiogenesis through its effects on vascular endothelial cells (ECs) and smooth muscle cells (SMCs), but the mechanisms coordinating this response are not well understood. Here we show th...
متن کاملMultifaceted Role of Neuropilins in the Immune System: Potential Targets for Immunotherapy
Neuropilins (NRPs) are non-tyrosine kinase cell surface glycoproteins expressed in all vertebrates and widely conserved across species. The two isoforms, such as neuropilin-1 (NRP1) and neuropilin-2 (NRP2), mainly act as coreceptors for class III Semaphorins and for members of the vascular endothelial growth factor family of molecules and are widely known for their role in a wide array of physi...
